Amidino-Rocaglates: A Potent Class of eIF4A Inhibitors

Amidino-Rocaglates: A Potent Class of eIF4A Inhibitors

Cell Chemical Biology
Volume 26, Issue 11, 21 November 2019, Pages 1586-1593

A screen of more than 200 rocaglates identified a novel, potent chemical series, amidino-rocaglates, that target eIF4A1 and eIF4A2 to potently inhibit translation and tumor cell viability, and are effective in synergizing with DNA-damaging agents in vivo against MYC-driven lymphomas. These compounds are promising anti-cancer agents.

The content in glucoputranjivin, which is the chemical marker for glucosinolates in S. officinale, has been determined using high-performance liquid chromatography (HPLC) and a pure reference standard. Glucoputranjivin and two isothiocyanates from S. officinale have been submitted to in vitro assays with the platform of bitter taste receptors of the T2Rs family. The results show that glucoputranjivin is a selective agonist of receptor T2R16.
